Tesla Motors repays Department of Energy loan nine years early

With a wire transfer of $451.8 million, Elon Musk has finally made good on his promise to repay the hefty loan Tesla received from the Department of Energy well ahead of its 2022 due date, beating even Musk's own five-year estimate. This comes on the heels of a very profitable first quarter, and is yet another sign the electric car maker is doing quite well on the greener side of things, despite a touch of bad press earlier this year. Anthony Weiner launches bid to become NYC mayor

NEW YORK (AP) ? Anthony Weiner's run for a renaissance is officially on.

The ex-congressman whose career imploded in a rash of raunchy tweets two years ago said in a YouTube video announcement late Tuesday that he's in the New York City mayoral race. He'd said last month he was considering it.

"I made some big mistakes and I know I let a lot of people down, but I also learned some tough lessons," he said in the video. "I'm running for mayor because I've been fighting for the middle class and those struggling to make it my entire life. And I hope I get a second chance."

With that, Weiner is embarking on an audacious comeback quest, hoping to go from punch line pol whose tweeted crotch shot was emblazoned on the nation's consciousness to leader of America's biggest city.

The video appeared late Tuesday but was not linked on his website or YouTube page. A call to Weiner was not immediately returned Wednesday.

The Democrat is jumping into a crowded field for September's primary. He's arriving with some significant advantages, including a $4.8 million campaign war chest, the possibility of more than $1 million more in public matching money, polls showing him ahead of all but one other Democrat ? and no end of name recognition.

His participation makes a runoff more likely, and many political observers feel he could at least get to the second round.

But Weiner also has continued to contend with questions about his character and the scandal that sank his career just two years ago.

After a photo of a man's bulging, underwear-clad groin appeared on his Twitter account in 2011, he initially claimed his account had been hacked. After more photos emerged ? including one of him bare-chested in his congressional office ? the married congressman eventually owned up to exchanging racy messages with several women, saying he'd never met any of them. He soon resigned.

In recent interviews, he has said he shouldn't have lied but did it because he wanted to keep the truth from his then-pregnant wife, Huma Abedin, a longtime aide to former Secretary of State Hillary Rodham Clinton. She told The New York Times Magazine that she has forgiven him.

Weiner has taken a series of steps recently to rehab his image and reintroduce himself, including the lengthy magazine profile and a series of local TV interviews. He hasn't responded to interview requests from The Associated Press.

He also has released a platform of sorts, a list of ideas styled as a blueprint for helping the city's middle class thrive. He's made a point of highlighting one or more of the concepts on most days, via his newly revived Twitter presence.

The suggestions, some of them updates from a mayoral run he nearly made in 2009, range from giving every public school student a Kindle reader to using Medicaid money to create a city-run, single-payer health system for the uninsured.

Some seem to draw on his Washington experience, such as making more use of a federal cigarette-smuggling law. But others fall squarely within City Hall, including suggestions to create a "nonprofit czar" in city government and eliminate paid positions for parent coordinators in schools.

The document also opens a window on a vision of the city ? a place with "a can-do attitude, competitive spirit and aggressive nature" ? that sounds not unlike Weiner himself. He was known during his seven terms in Washington as a vigorous defender of Democratic viewpoints, unafraid to get combative whether it was on cable TV or the House floor, and as a tireless and instinctive politician.

"Anybody who underestimates Anthony Weiner's ambition is a fool. And anybody who underestimates his ability as a candidate is a fool," retired Hunter College political science professor Kenneth Sherrill said. But "we're going to see, basically, if Weiner can take hits as well as he can dish them out."

In seeking a second chance from the public, Weiner will have to overcome some voters' misgivings. In a recent NBC New York-Marist Poll, half said they wouldn't even consider him, though the survey also showed that more registered Democrats now have a favorable than unfavorable impression of him.

Weiner can expect opponents to hammer at his prior prevaricating, and he said in a recent interview on the RNN cable network that he couldn't guarantee that no more pictures or people would emerge.

And while he might welcome attention to his policies rather than his past, they also have attracted some criticism. About a dozen young people recently demonstrated outside his Manhattan apartment building to denounce his proposal to make it easier to suspend disruptive public school students; "(hash)Weiner: You ask for a second chance in (hash)NYC2013 but deny students a second chance," read one sign, using Twitter's beloved hashtag marks.

Since leaving office, Weiner has put his government experience to work as a consultant for various companies.

His Democratic opponents include City Councilman Sal Albanese; Public Advocate Bill de Blasio; Comptroller John Liu; City Council Speaker Christine Quinn; the Rev. Erick Salgado, a pastor; and former Comptroller Bill Thompson.

Republican contenders include billionaire businessman John Catsimatidis, former Metropolitan Transportation Authority Chairman Joseph Lhota and homelessness-aid organization head George McDonald. Former White House housing official Aldolfo Carrion Jr., a Democrat who recently dropped his party affiliation, is running on the Independence Party line and also interested in the Republican nomination.

In a commencement address at Atlanta's historically black Morehouse College, President Obama said graduates should "find time to defend the powerless."

The president said his own success was due to "the special obligation I felt, as a black man like you, to help those who need it most, people who didn't have the opportunities that I had ? because there but for the grace of God, go I. Kremlin orders Russian pollster to register itself as 'spy'

Russia's only independent polling agency, the Levada Center, may face closure after Russian prosecutors ordered it to register as a "foreign agent" ? a term that's synonymous with "spy" in Russian ? under a new law designed to clamp down on nongovernmental organizations that receive any amount of funding from abroad and engage in any form of activity that authorities deem political.

The prosecutor's warning, delivered to the Levada Center on Monday and reproduced on the agency's website, essentially declares that Levada's world-renowned public opinion research is not considered by Russian authorities to be scientific work but rather "political activity" that is "aimed at shaping public opinion on government policy."

Levada, a registered NGO, is the only one of three major Russian pollsters that is independent of state funding and management. It has often had a tense relationship with the Kremlin. But many experts expressed surprise Monday over the prosecutor's threat, if only because Levada's sociological research ? including regular tracking polls on Vladimir Putin's public popularity ? has always been highly regarded on all sides of the political spectrum and is constantly cited by officials, parliamentarians, and state-run media.

In a statement, Levada's director Lev Gudkov said the order to cease publishing its work until it registers as a "foreign agent" puts the agency in "an extremely difficult situation."

"The prosecutors office is hanging our organization on the hook of possible fines while at the same time undermining its credibility and business reputation. Basically, the extremely vague definitions of 'political activity' and 'foreign funding' being applied here allow for the most arbitrary and elastic interpretations.... Hence, we face the possibility of administrative action against the management of our agency and even possible liquidation," he said.

Dozens of Russian NGOs have already been ordered by prosecutors to wear the "foreign agent" label in all their public activities, or face crippling fines and, ultimately, police shutdown. None have yet complied with the order, which they say does nothing to improve financial transparency and is solely designed to destroy their credibility with the Russian public, official institutions, and other organizations by saddling them with a label that means "spy" and nothing else in Russian.

Among those that have been singled out by prosecutors are the grassroots election monitoring organization Golos, the corruption watchdog Transparency International, and human rights monitors such as Memorial.

All of the targeted groups insist that they do not engage in partisan political activities of any kind, but that prosecutors are deploying a definition of "political" that includes any activities that might irritate authorities.

"This is not an attack against us specially but it is part of a growing assault against all NGOs and active civil society," says Alexei Grazhdankin, Levada's deputy director.

"What authorities want to do with Levada is to regulate the way we do our sociological work and control our relations with clients.... We are not engaged in political activity because we do not interfere, we just track the changes in public opinion and record them. It's not clear to us how this law is being applied, or how the publication of our data can be interpreted as a means to influence public opinion," he says.

"When some of our polls about Putin or the government get published, they get angry at us. We do other polls about the protest movement and the opposition that get them upset. We're just doing our job, which is to try to shed light on what's happening in the country," he adds.

The prosecutor's warning letter alleges that Levada has received $777,000 over the past four years in funding from outside groups such as the MacArthur Foundation, the Ford Foundation, and George Soros? OSI Assistance Foundation.

The agency also earns money by doing sociological research and market surveys on behalf of foreign clients, Mr. Grazhdankin says.

In his statement, Mr. Gudkov insists that such funding has not amounted to more than 3 percent of Levada's budget in recent years.

"Unlike other polling organizations, we have no direct state financing or state grants for carrying out surveys that often entail significant financial and organizational expenses," writes Gudkov. "The funds we have received from foreign sources (including donations and grants won on a competitive basis) or payment for ordered surveys by foreign organizations (such as universities, mass-media, universities and consulting firms) makes up an insignificant part of the budget of Levada Center, basically between 1.5 percent and 3 percent in various years," he added.

All major NGOs have agreed among themselves to refuse prosecutors' orders to don the "foreign agent" label, which they argue is tantamount to swallowing a poison pill.

The authorities' next step, many analysts argue, will be to step up the pressure with huge fines until the biggest NGOs buckle, or send in police to close them down.

"In a sense we are returning to Soviet times, when all political information will be generated and doled out by the authorities," says Dmitry Oreshkin, head of the Mercator Group, an independent Moscow media consultancy.

"It's a convenient situation for the authorities, who can just write down the number of votes they need and it will be so reported. Economic indexes will grow constantly, and support for our beloved leader will be always buoyant. And it will go on that way until everything once again collapses," he says.

Mechanism linking key inflammatory marker to cancer identified

May 20, 2013 ? In a new study described in the journal Oncogene, researchers reveal how a key player in cell growth, immunity and the inflammatory response can be transformed into a primary contributor to tumor growth.

Scientists call this Jekyll-and-Hyde molecule NF-kappa B. In healthy cells, it is a powerful "first responder," a vital part of the body's immune and inflammatory responses. It spends most of its life in the cell's cytoplasm, quietly awaiting orders. But when extracellular signals -- of a viral or bacterial invasion, for example -- set off chemical alarms, the cell unchains this warhorse, allowing it to go into the nucleus where it spurs a flurry of defensive activity, including the transcription of genes that trigger inflammation, promote cell proliferation and undermine cell death.

Researchers have known for years that a hyperactive form of NF-kappa B that gets into the nucleus and stays there is associated with various cancers. But they didn't know what was keeping it active in the nucleus.

"Normally in the cell NF-kappa B is in the cytosol, it's not in the nucleus, and it's not activated," said University of Illinois medical biochemistry professor Lin-Feng Chen, who led the new study. "You have to stimulate normal cells to see NF-kappa B in the nucleus. But in cancer cells without any stimulation you can see this nuclear form of NF-kappa B. The cell just won't die because of this. That is why NF-kappa B is so important in cancer."

In the new study, Chen's group found that another molecule known to help regulate gene expression, called BRD4, recognizes a specific amino acid on a subunit of the NF-kappa B protein complex after the amino acid has been marked with a specific tag, called an acetyl group. This "acetylation" allows the BRD4 to bind to NF-kappa B, activating it and preventing its degradation in cancer cells.

Previous studies had shown that BRD4's recognition of the acetylated subunit increased NF-kappa B activation, but this recognition had not been linked to cancer.

BRD4 belongs to a class of molecules that can recognize chemical markers on other proteins and interact with them to spur the marked proteins to perform new tasks. Chemical "readers" such as BRD4 are important players in the field of epigenetics, which focuses on how specific genes are regulated.

"In epigenetics, there are writers, there are readers and there are erasers," Chen said. The writers make modifications to proteins after they are formed, without changing the underlying sequence of the gene that codes for them. These modifications (such as acetylation) signal other molecules (the readers) to engage with the marked proteins in various ways, allowing the proteins to fulfill new roles in the life of the cell. Epigenetic erasers remove the marks when they are no longer of use.

Such protein modifications "have been shown to be critically involved in transcription regulation and cancer development," the researchers report.

To test whether BRD4 was contributing to the sustained presence of NF-kappa B in the nucleus of cancer cells, Chen and his colleagues exposed lung cancer cells in cell culture and in immune-deficient mice to JQ1, a drug that interferes with BRD4 activity. Exposure to JQ1 blocked the interaction of BRD4 and NF-kappa B, blocked the expression of genes regulated by NF-kappa B, reduced proliferation of lung cancer cells and suppressed the ability of lung cancer cells to induce tumors in immune-deficient mice, the researchers found.

The researchers also discovered that depletion of BRD4 or the treatment of cells with JQ1 induced the degradation of the NF-kappa B subunit recognized by BRD4.

Chen said that BRD4 likely prevents other molecules from recognizing the hyperactive NF-kappa B in the nucleus and marking it for degradation.

"This is an example of how epigenetic regulators and NF-kappa B may one day be targeted for the treatment of cancer," he said.

Researchers from Illinois biochemistry professor Satish Nair's laboratory and from the laboratory of James Bradner at the Dana-Farber Cancer Institute contributed to this study.
